Dissertation note | The study was conducted with objective of optimized xylanase using local raw materials from indigenous isolate of Apergillus jlavus. The fungus was grown on different carbon sources including wheat bran, rice polishing for the production of xylanase enzyme. All four fungi produced xylanase activity in the medium containing wheat bran and rice polishing (1%) at pH 7.0 for 4 days. Maximum activity (14.3U/mL) was depicted by A. jlavus3 in medium with wheat bran. On the basis of better xylanase production A. jlavus3 was selected for further production and characterization of enzyme studies. The highest xylanase activity was achieved in cultivation with wheat bran (lS.8U/mL). A slightly higher quantity of xylanase was produced by the strain in wheat bran-supplemented medium (18.5 U/mL) followed by rice polishing (17.9 U/mL) when 3% carbon sources were used. The effect of supplementation of different source of nitrogen on xylanase activity by A. flavus was studied with 3 % carbon source. Of all the nitrogen sources investigated, yeast extract (organic source) was the most promising and the corresponding xylanase production was 19.9 UlmL (wheat bran) and 18.3 U/mL (rice polishing). Com step liquior was used to enhance the activity of xylanase which was approx. 10 % higher than that of control medium lacking com step liquior. The highest level of xylanase activity as well as extracellular protein using wheat bran was reported .Maximum xylanase production occurred at pH 5.5 and activities of enzyme were obtained at temperature 30°C for A. jlavus. The enzyme was purified by gel filtration, ion exchange chromatography. The enzyme activity was characterized on different temperatures and pH ranges. |
